RESUMO

Gelidocalamusalbozonatus W. G. Zhang, S. R. Yi & Y. L. Li, a new species of Gelidocalamus, collected from Pengshui County of Chongqing City in China, was described and illustrated herein. In this study, key morphological characters were compared between the new species and other eight "gelido-" members of Gelidocalamus. By using scanning electron microscopy (SEM), its leaf epidermal characters were observed in comparison with those of another three Gelidocalamus representatives. Our results show that the new taxon has the typical characteristics of the genus Gelidocalamus, both macromorphologically and micromorphologically. Moreover, it was most similar to G.tessellatus, but differed by a ring of white tomenta below per node, culm sheath base with densely purple verrucous setae and foliage leaf blades mesophyll.

The genus Gelidocalamus T. H. Wen, endemic to southern China, is a small but taxonomically problematic genus of Arundinarieae (Poaceae, Bambusoideae). During field work, a population of Gelidocalamus from Zixing, Hunan, was discovered, appearing to be distinct from our previously identified collection. Comparisons of the population of Zixing were performed by using scanning electron microscopy (SEM) and a plastid genome-based phylogeny. Morphologically, it was mostly similar to G.multifolius, but differed by culm leaf erect with densely white pubescence, apical branch sheath much longer than the internodes and foliage leaf larger. Phylogenetically, the new species was well-supported as a sister to the clade of G.multifolius + G.tessellatus, and the above three taxa were clustered in the Shibataea clade (IV) of Arundinarieae. Thus, the new species, formally named as Gelidocalamuszixingensis W.G.Zhang, G.Y.Yang & C.K.Wang, was described and illustrated herein.

BACKGROUND: The purpose of this study was to retrospectively analyze clinical characteristics and prognostic risk factors of urosepsis patients admitted to two intensive care units in Shanghai, China. METHODS: Clinical data from patients diagnosed with urosepsis were retrospectively retrieved and analyzed from ICU in two regional medical centers from January 2015 to December 2019. RESULTS: Two hundred two patients were included in the subsequent analysis eventually, with an average age of 72.02 ± 9.66 years, 79.21% of the patients were female and the mortality rate of 15.84%.The proportion of patients with chronic underlying diseases such as diabetes and hypertension was relatively high (56.44, 49.50%, respectively), and the incidence of shock was also high (41.58%) correspondingly. The most common pathogen isolated was Escherichia coli (79.20%), of which the extended-spectrumß-lactamases (ESBLs)(+) accounted for 42.57%. In multivariate analysis, the strongest predictors for death were mechanical ventilation (OR 7.260, 95% CI 2.200-23.963; P = 0.001),chronic kidney disease (CKD) (OR 5.140, 95% CI 1.596-16.550; P = 0.006), APACHE II score (OR 1.321, 95% CI 1.184-1.473; P < 0.001) and lactate (OR 1.258, 95% CI 1.037-1.527; P = 0.020). Both APACHE II score and lactate had the ideal predictive value, with the area under the ROC curve (AUC) of 0.858 and 0.805 respectively. CONCLUSION: The patients with urosepsis were characterized by a higher proportion of female, older age, more percentage of comorbidities in this region, and patients with ESBLs (+) Escherichia coli infection were more prone to shock. Mechanical ventilation, comorbidity with CKD, APACHE II score and lactate were independent risk factors for death in urosepsis patient, but lactate level and APACHE II score had better predictive value for prognosis.

Phyllostachys edulis (Bambusoideae) is a temperate woody bamboo with a long history of cultivation in China. Phyllostachys edulis f. curviculmis is the latest new forma that repored in 2018. Here, we performed the complete chloroplast genomes of P. edulis and P. edulis f. curviculmis using genome skimming. The length of two chloroplast genomes was 139,678 bp, and their GC contents were 38.9%. The sequences of each species contained 132 unique genes, including 39 tRNA, eight rRNA, and 85 protein-coding genes. Moreover, in subspecies-level, P. edulis 'Pachyloen' and P. edulis f. curviculmis are identical to P. edulis in the terms of chloroplast genome size, structure, and composition, further indicating their affinity.

The complete chloroplast genome of a rare deciduous tree species with ornamental value, Sinomanglietia glauca, was first determined. It was 160,170 bp in length, including a pair of inverted repeat (IR, 26,567 bp) regions separated by a small single copy (SSC, 18,842 bp) sequence and a large single copy (LSC, 88,194 bp) sequence. The chloroplast genome contained 132 genes, consisting of 87 CDS, 8 rRNA genes, and 37 tRNA genes. Thirty-four SSR sites were detected in the chloroplast genome. The phylogenetic analysis revealed that all sampled Manglietia species were clustered together, and S. glauca was placed as sister to the clade Manglietia, indicated that the genus Sinomanglietia may be legitimate and should be recovered.

The complete chloroplast genome sequence of Gelidocalamus xunwuensis, firstly determined here, is 139,705 bp in length, inclusive of a pair of inverted repeat (IR, 21,817 bp) regions separated by a small single copy (SSC, 12,803 bp) and a large single copy (LSC, 83,268 bp). It contains 132 genes, such as 85 CDS, 8 rRNA genes, and 39 tRNA genes, respectively. The phylogenetic analysis shows that G. xunwuensis is highly clustered in the shibataea clade (III) of Arundinarieae, sister to the clade of G. tessellatus + Ferrocalamus rimosivaginus.

The genus Phyllostachys is economically important; however, only a small amount of complete plastid genomes have been reported to date. Here, we characterized two complete chloroplast genomes of Phyllostachys using genome skimming. The chloroplast genomes of Phyllostachys reticulata and Phyllostachys edulis 'Pachyloen' were 136,689 bp and 139,678 bp in length, respectively, and their GC contents were 38.8% and 38.9%, respectively. The sequences of each species contained 132 unique genes, including 39 tRNA, eight rRNA, and 85 protein-coding genes. Phylogenetic analysis shows that all selected Phyllostachys species were grouped into one well-supported clade in the Phyllostachys clade (V) of Arundinarieae. Moreover, in terms of chloroplast genome size, structure, and composition, P. edulis 'Pachyloen' is identical to P. edulis, further indicating the affinity between them.

RESUMO

Gelidocalamus xunwuensis W.G.Zhang & G.Y.Yang, a new species collected from Xunwu County of Jiangxi Province in China, is described and illustrated. The new species is similar to G. stellatus in the habit, but differs by internodes sparsely hairy with granuliferous warts, culm sheath stiffly hairy, culm sheath blade broadly lanceolate to narrowly triangular, each node with a ring of appressed trichomes below, foliage leaves broadly lanceolate to narrowly oblong, and new shoots occurring in late October.

Bamboo (Phyllostachys pubescens) expansion into adjacent forests is a widespread phenomenon in subtropical regions, and it has greatly changed the dominance hierarchy from trees to bamboos. This process may be accompanied by changes in productivity, nutrients accumulation and biogeochemical cycles. We compared the net primary production (NPP) and major pools and fluxes of nitrogen (N) in bamboo-dominant forest (BDF) and neighboring secondary evergreen broadleaved forest (EBF) in South China using the space-for-time substitution method. We found that the mean NPP of the BDF was 30.0 t ha-1 yr-1, which was 51.5% greater than that of the EBF (19.8 t ha-1 yr-1). The plant N pool for the BDF was 37.5% larger than that of the EBF, whereas the soil inorganic N pool significantly decreased by 31.2% with conversion of the EBF to BDF. Additionally, the ratio of N return to N uptake was 0.69 in the BDF and 0.88 in the EBF because of the lower litter N return of the BDF compared with that of the EBF. These results indicated that the expansion of P. pubescens significantly increased the NPP and plant N accumulation but reduced the soil N available pool and slowed the N cycling rate, which could lead to soil degradation.

The effects of Phyllostachys edulis and Oligostachyum oedogonatum expansion on species diversity of broad-leaved forests were investigated in Wuyishan National Nature Reserve, Jiangxi Province, China. Ph. edulis and/or O. oedogonatum expansion changed community structure and species composition. The co expansion of the two bamboos in high intensity (Ph. edulis was 30-50 ind·100 m-2, O. oedogonatum was 300-500 ind·100 m-2) changed the density ratio of arbor and shrub from 2:8 to 1:9, and density ratio of bamboo and tree from 0:10 to 9:1. The main effects of Ph. edulis and O. oedogonatum on the species diversity were significant. Ph. edulis mainly influe-nced arbor layer, while O. oedogonatum influenced shrub layer more, and the interaction effect of two bamboos was not remarkable. There existed addition effects between Ph. edulis and O. oedogonatum on species diversity. The co-occurrence of two bamboos in high intensity decreased the Shannon index of community by 91.3%. Ph. edulis and O. oedogonatum did not compete obviously unless in high density. It was addition effect rather than interaction effect that changed the community structure and reduced species diversity when broad leaved forest suffered invasion by Ph. edulis and O. oedogonatum.

To detect the ecological process of the succession series of Phyllostachys glauca forest in a limestone mountain, five niche models, i.e., broken stick model (BSM), niche preemption model (NPM), dominance preemption model (DPM), random assortment model (RAM) and overlap- ping niche model (ONM) were employed to describe the species-abundance distribution patterns (SDPs) of 15 samples. χ² test and Akaike information criterion (AIC) were used to test the fitting effects of the five models. The results showed that the optimal SDP models for P. glauca forest, bamboo-broadleaved mixed forest and broadleaved forest were DPM (χ² = 35.86, AIC = -69.77), NPM (χ² = 1.60, AIC = -94.68) and NPM (χ² = 0.35, AIC = -364.61), respectively. BSM also well fitted the SDP of bamboo-broadleaved mixed forest and broad-leaved forest, while it was unsuitable to describe the SDP of P. glauca forest. The fittings of RAM and ONM in the three forest types were all rejected by the χ² test and AIC. With the development of community succession from P. glauca forest to broadleaved forest, the species richness and evenness increased, and the optimal SDP model changed from DPM to NPM. It was inferred that the change of ecological process from habitat filtration to interspecific competition was the main driving force of the forest succession. The results also indicated that the application of multiple SDP models and test methods would be beneficial to select the best model and deeply understand the ecological process of community succession.

AIM: To investigate the reciprocal modulation between microRNA (miRNA) and DNA methylation via exploring the correlation between miR-373 and methyl-CpG-binding domain protein (MBD)2. METHODS: MiR-373 expression was examined using the TaqMan miRNA assay. Methylation of miR-373 was investigated using methylation-specific polymerase chain reaction, and recruitment of methyl binding proteins was studied using the chromatin immunoprecipitation assay. Mutation analysis was conducted using the QuikChange™ Site-Directed Mutagenesis kit. The activity of miR-373 gene promoter constructs and targeting at MBD2-three prime untranslated region (3'UTR) by miR-373 were evaluated by a dual-luciferase reporter gene assay. RESULTS: In hilar cholangiocarcinoma, miR-373 decreased and was closely associated with poor cell differentiation, advanced clinical stage, and shorter survival. The promoter-associated CpG island of miR-373 gene was hypermethylated and inhibited expression of miR-373. MBD2 was up-regulated and enriched at the promoter-associated CpG island of miR-373. Methylation-mediated suppression of miR-373 required MBD2 enrichment at the promoter-associated CpG island, and miR-373 negatively regulated MBD2 expression through targeting the 3'UTR. CONCLUSION: MiR-373 behaves as a direct transcriptional target and negative regulator of MBD2 activity through a feedback loop of CpG island methylation.
